Hepatoprotective efficacy of aqueous acetone extract of flowers of Clerodendrum infortunatum on acetaminophen induced toxicity in HepG2 cells
Abstract
The present study was aimed at investigating the hepatoprotective efficacy of
aqueous acetone extract of flowers of Clerodendrum infortunatum on acetaminophen
induced toxicity in HepG2 cells.The hepatoprotective effect of extracts on acetaminophen
induced toxicity was determined by adding 20 mM acetaminophen followed by extracts at
final concentrations of 6.25 µg/ml, 12.5 µg/ml, 25 µg/ml, 50 µg/ml and100 µg/ml from a
stock of 1 mg/ml .The % difference in viability was determined by standard MTT assay
after 24 hours of incubation. Among five different concentrations used in the study (50 to
800 mg/ml), acetone extract showed maximum scavenging activity. Addition of
acetaminophen at 20 mM concentration reduced the cell viability to 35% with significant
changes in cell morphology. Addition of extracts produced betterment in cell viability
and morphology. 10 µg/ml of extract was found to ameliorate the hepatotoxicity and
increased the cell viability to 87.9 % which could be considered significant.
Phytochemical screening of this plant revealed the presence of steroids, flavonoids,
phenolics and alkaloids which could be responsible for the possible hepatprotective
action
